Solid phase extraction (SPE) is a physical extraction process that includes liquid and solid phases. In the extraction process, the adsorption force of the solid to the analyte is greater than the sample mother liquor. When the sample passes through the SPE column, the analyte is adsorbed on the solid surface, and other components pass through the column with the sample mother liquor. Finally, the analyte is removed with an appropriate solvent Eluted. SPE has a wide range of applications, such as the analysis of biological fluids including blood, urine, serum, plasma and cytoplasm; the analysis of milk processing, wine, beverages and fruit juices; the analysis and monitoring of water resources; fruits, vegetables, grains, and various plant tissues Animal tissues; solid medicines such as tablets. Analysis of pesticide and herbicide residues in fruits, vegetables and foods, analysis of antibiotics and clinical drugs, etc.
(1) Carefully take out the solid phase extraction device and place it gently on the workbench.
(2) Carefully take out the upper cover of the SPE device (handle gently so as not to damage the small tube), insert the standard test tube into the hole of the partition in the vacuum chamber, and then cover the upper dry cover, and ensure that the cover is guided downwards. The flow tube and the test tube correspond one by one, and the square sealing ring of the cover plate has a good sealing performance with the vacuum chamber. If it is not easy to seal, it can be tightened with a rubber band to increase the tightness.
(3) If you have purchased an independent adjustment, you must first insert the adjustment valve into the extraction hole of the cover;
(4) If you do not need to do 12 or 24 samples at a time, plug the needle tube tight valve into the unused extraction hole;
(5) If an independent control valve is purchased, turn the control valve knob of the unused extraction hole to the horizontal sealing state;
(6) Insert the solid phase extraction cartridge into the extraction hole or valve hole of the upper cover (turn the regulating valve knob to the upright open state); connect the extraction device and the vacuum pump with a hose, and tighten the pressure regulating valve;
(7) Inject the samples or reagents to be extracted into the extraction column, and start the vacuum pump, then the sample in the extraction column will flow through the extraction column to the test tube below under the action of negative pressure. At this time, the flow rate of the liquid can be adjusted and controlled by adjusting the pressure reducing valve.
(8) After the liquid in the needle tube is pumped, turn off the vacuum pump, unplug the enrichment column from the device, remove the upper cover of the device, take out the test tube and pour it out.
(9) If you don’t want to use the test tube to connect the liquid, you can take out the test tube rack, put it into a container of suitable size, and take it out after the first extraction.
(10) Put the clean test tube into the device, close the cover, insert the SPE cartridge, add the required extraction solvent to the needle tube, start the vacuum pump, turn off the power after the liquid is drained, and take out the test tube for use. The extraction and sample preparation are complete.
(11) Put the test tube into a nitrogen drying apparatus and purify and concentrate with nitrogen, and the preparation is complete.
(12) Dispose of the solvent in the test tube, and rinse the test tube for reuse.
(13) In order to save the cost of using the SPE column, after each use, the SPE column should be rinsed with eluent to ensure the properties of its packing.
